چکیده

microRNAs (miRNA,miR)playan important role in cancer cell growthandmigration;however, thepotential roles of miRNAs in osteosarcoma remain largely uncharacterized. By applying a miRNAmicroarray platform and unsupervised hierarchical clustering analysis, we found that several miRNAs have altered expression levels in osteosarcoma cell lines and tumor tissues when compared with normal human osteoblasts. Three miRNAs, miR-199a-3p, miR-127-3p, and miR-376c, were significantly decreased in osteosarcoma cell lines, whereas miR-151-3p and miR-191 were increased in osteosarcoma cell lines in comparison with osteoblasts. Transfection of precursor miR-199a-3p into osteosarcoma cell lines significantly decreased cell growth and migration, thus indicating that the inhibition effect is associatedwith an increase in theG1-phase and a decrease of the S-phase cell population. In addition, we observed decreasedmTOR and Stat3 expression in miR-199a-3p transfected cells. This study provides new insights formiRNAs in osteosarcoma and suggests thatmiR-199a-3p mayplay a functional role in osteosarcoma cell growth andproliferation. RestoringmiR-199a-3p’s functionmay provide therapeutic benefits in osteosarcoma. Mol Cancer Ther; 10(8); 1337–45. 2011 AACR.

MicroRNA-199a-3p is downregulated in gastric carcinomas and modulates cell proliferation.

MicroRNAs (miRNAs) are small non-coding RNAs that regulate the translation of targeted mRNAs. An increasing amount of evidence indicates that miRNAs play important role in cancer pathogenesis, apoptosis, proliferation, and differentiation as oncogenes or tumor suppressors. Recently, miRNA-199a has been shown to be involved in many human cancers, although the role of miRNA-199a-3p in gastric can...

MicroRNA-199a-3p and microRNA-34a regulate apoptosis in human osteosarcoma cells

miRNAs (microRNAs) are small non-coding RNAs [18-25 nt (nucleotides)] that regulate gene expression mainly through affecting post-transcriptional modification. Osteosarcoma is an aggressive sarcoma of the bone characterized by a high level of genetic instability and recurrent DNA deletions and amplifications. miRNAs play an important role in cancer cell growth and migration; however, the potent...

CD44 is a direct target of miR-199a-3p and contributes to aggressive progression in osteosarcoma

Osteosarcoma is the most common primary bone malignancy in children and adolescents. Herein, we investigated the role of cluster of differentiation 44 (CD44), a cell-surface glycoprotein involved in cell-cell interactions, cell adhesion, and migration in osteosarcoma. We constructed a human osteosarcoma tissue microarray with 114 patient tumor specimens, including tumor tissues from primary, me...

MiR-199a-3p regulates mTOR and c-Met to influence the doxorubicin sensitivity of human hepatocarcinoma cells.

MicroRNAs (miRNA) have rapidly emerged as modulators of gene expression in cancer in which they may have great diagnostic and therapeutic import. MicroRNA-199a-3p (miR-199a-3p) is downregulated in several human malignancies including hepatocellular carcinoma (HCC). Here, we show that miR-199a-3p targets mammalian target of rapamycin (mTOR) and c-Met in HCC cells. Restoring attenuated levels of ...
